ETAS® is a patented, enzyme-processed extract from asparagus rhizomes with stress-reducing and sleep-improving properties. This plant extract, produced through a proprietary process from the lower, woody part of the asparagus (Asparagus officinalis), possesses stress-reducing and sleep-enhancing properties.

ETAS® is the first extract in the world whose mechanism of action involves triggering the release of Heat Shock Protein 70 (HSP70), a protein usually produced by the body in response to stress related to excessive heat exposure. This effect is similar to the benefits of hot spring baths, which delay aging processes and relieve stress. ETAS® also demonstrates other beneficial effects, such as improving heart rate variability, enhancing cognitive abilities, and reducing fatigue. ETAS® is rich in compounds known as hydroxymethylfurfural derivatives, identified as novel compounds capable of inducing HSP70 expression (Ito, 2013). One such compound found in ETAS® is a derivative known as asfural (a derivative of 5-hydroxymethyl-2-furfural – HMF-1). It shows significant activity in increasing HSP70 mRNA expression and a range of biological activities, including antioxidant effects, reduction of myocardial ischemia, and improvement of blood flow (Ito, 2013).

Tender asparagus buds are a healthy and nutritious food often used to prepare exquisite dishes, while the woody parts of the stems are typically discarded. Japanese scientists discovered that after appropriate processing with enzymes and extracting elements from the discarded part of the rhizome, it exhibits extraordinary health properties.

How does ETAS® reduce stress parameters?

Heat shock proteins (HSPs) are intracellular proteins produced in response to various stress factors, mainly excessive heat exposure. They help protect and repair the body’s cellular proteins from thermal damage, preventing them from becoming misfolded and denatured, which would cause them to lose their function. There is a hypothesis that these proteins are responsible for the anti-stress and anti-aging effects of thermal and spa treatments. The production of heat shock proteins is also stimulated by exercise and calorie restriction, which is known from animal studies to extend lifespan (Iguchi, 2012, Calderwood, 2009). Some HSPs (HSP70) directly protect cells from damage that could lead to premature cell death, known as apoptosis. However, the heat shock response declines with age, and the weakening of this response contributes to aging by allowing the development of protein aggregation diseases (e.g., Alzheimer’s disease). This leads to reduced cellular viability and decreased longevity (Calderwood, 2009). HSP70 (with a molecular weight of 70,000) triggers a range of beneficial physiological effects in the body, including reducing cell mortality, exerting anti-inflammatory and antioxidant effects. The formation of this protein is stimulated by several factors, such as heat, hunger, alcohol consumption, and ultraviolet radiation (Doeppner, 2013; Matsuda, 2013; Agaki, 2013). It has been found that HSP70 protein levels also decrease with age, which may be associated with a low-grade inflammation accompanying the aging process (Njemini, 2011).

Heat shock protein information – research results involving ETAS®

In studies on sleep-deprived mice, ETAS® clearly inhibits the secretion of corticosterone, a stress hormone. Human studies have shown that ETAS® can reduce stress and improve sleep quality. In a study involving 14 healthy volunteers divided into 2 groups, a placebo group and an ETAS® group (7 people in each group), the ETAS® group consuming 300 mg of ETAS® daily for a week showed a threefold increase in HSP70 protein expression at the mRNA level in white blood cells after consumption compared to before consumption (Amino Up, sales bulletin, Hokkaido, Japan). In a clinical study involving humans, stress was reduced as HSP70 protein levels in peripheral blood increased due to ETAS® consumption (Nishira). Another method of studying the impact of ETAS® on stress reduction was observing its effects on the autonomic nervous system (ANS). The observation was based on heart rate variability (HRV) analysis by measuring pulse wave acceleration using a Plus TAS9 pulse analyzer. In a clinical study involving 30 healthy volunteers divided into 2 groups, a placebo group and an ETAS® group, with 15 people in each group, each group took 400 mg of ETAS® or a placebo daily for 4 weeks. The study results showed that ETAS® improved stress response by enhancing ANS function.

Healthier, longer, and deeper sleep with the versatile ETAS® supplement

In the group taking ETAS®, sleep-related issues decreased, as demonstrated in a randomized, double-blind, placebo-controlled clinical trial conducted at the University of Information Science in Hokkaido. Sixteen healthy male volunteers with sleep problems received 300 mg/day of ETAS®. Serum cortisol levels increased by 45% in the placebo group, while in the ETAS® group, the increase was only 10% (Amino Up, unpublished study). In the ETAS® group, the increase in cortisol-induced stress in the blood was inhibited by about 80%. In the ETAS® group, salivary cortisol levels increased by only 47% compared to 75% in the control group. In the ETAS® group, the psychological stress marker known as chromogranin A decreased 2.5 times more than in the control group.

To assess the impact on sleep quality and determine whether ETAS® would be effective in preventing sleep disorders, a clinical trial in humans used an actigraph analyzer. Actigraphy allows monitoring of rest and activity cycles during sleep using a sensor placed on the patient’s body. Taking ETAS®, which was safe and well-tolerated, resulted in significant improvement in sleep quality. Serum cortisol, salivary cortisol, and chromogranin A levels decreased, indicating improved stress levels. ETAS® was effective in improving sleep quality and reducing psychological stress (Nishira). In another placebo-controlled study evaluating the impact of ETAS® on sleep quality improvement, the Athens Insomnia Scale was used to assess sleep onset, nighttime and early-morning awakenings, sleep duration, sleep quality, frequency and duration of disturbances, stress caused by experiencing insomnia, and daily functioning disruptions. The ETAS® group scored 0.40 percentage points higher than the placebo group. Sleep quality and frequency of dreams decreased by nearly 2 points in the placebo group, but increased by 5.4 points in the ETAS® group.

Mood improvement and energy boost – feel better every day with ETAS®

ETAS® effectively reduces fatigue and dysphoria (unhappiness) caused by stress, even with a daily dose of 150 mg (Waki, 2013). In a randomized, double-blind, placebo-controlled crossover trial published in Clinical Nutrition, 25 healthy volunteers received 150 mg/day of ETAS® or placebo for 28 days, followed by a 14-day washout period. The study used a questionnaire asking participants about their energy levels or feelings of fatigue while sitting, and a memory calculation task was used to induce psychological stress. Autonomic nervous functions were measured using heart rate variability analysis with an active orthostatic test. After the memory calculation task, various stress parameters, including serum catecholamine hormone levels, salivary sIgA concentration (an immune factor in saliva that decreases due to stress), and cortisol levels, were analyzed. It was found that after 28 days of using ETAS®, sIgA levels increased, feelings of daily fatigue decreased, and dysphoria was reduced.

Other beneficial effects of ETAS® on physiological functions

ETAS® has strong, positive effects on various areas of physiological function. It has anti-inflammatory effects in liver cells by inhibiting nitric oxide production (Nishizawa, et al.). In intestinal epithelial cells, ETAS® reduces cell toxicity and excessive cell proliferation under normal conditions and oxidative stress. Adding ETAS® to neuronal cell cultures alleviates cellular stress by increasing HSP70 protein expression. In studies on Alzheimer’s disease, it was found that pretreating PC12 nerve cells with ETAS® helped prevent cell death induced by beta-amyloid (βA). ETAS® significantly reduced DNA damage caused by βA, as well as decreased the production of reactive oxygen species. Study results showed that ETAS® has specific properties in protecting brain cells from damage by increasing the viability of cells exposed to beta-amyloid (Ogasawara).

HSP – heat shock proteins maintain proper daily cellular homeostasis, protecting their proteins from damage and restoring their proper function. HSP expression is induced to protect cells from stressors. However, the HSP response to stressors decreases with age.

Misfolded protein – a properly functioning protein should be folded. HSP repairs misfolded proteins or degrades them when the protein is irreparable.
